Summary, etc. A study on “Embryo Rescue in the Interspecific Crosses of Abelmoschus” was undertaken to overcome the problem of embryo abortion in the interspecific crosses of Abelmoschus. Hybridization was attempted with A. esculentus var. Kiran and A. esculentus var. Anakomban (representing the genotypes susceptible to YVM) and A. moschatus, A. tetraphyllus and A. manihot (representing the genotypes resistant to YVM).
The fruit set was low (28.00 – 92.00%) in all the interspecific crosses compared to the female parent (81.00 – 97.81 %). Crosses with A. esculentus var. Kiran or A. esculentus var. Anakomban as female parent recorded higher fruit set (71.05 – 92.00%) than those with wild or semi – wild species as female parent (28.00 – 71.67%).
The crossed fruits resembled the fruits of female parent in morphology irrespective of the pollen source. Fruit weight, number of seeds per fruit and 100 seed weight were reduced in the interspecific crosses.
All crossed seeds showed good viability except in crosses involving A. moschatus. The seeds in crosses involving A. moschatus were shrivelled and non – viable.
Experiments on in vitro embryo rescue, to overcome the post – zygotic incompatability, revealed that culturing 12 and 15 day old embryos of A. esculentus var. Kiran x A. moschatus and A. esculentus var. Anakomban x A. moschatus on MS medium supplemented with BA 0.5 mg 1-1 and CW 150 ml 1-1 yielded transplantable seedlings.
Embryos of crosses A. moschatus x A. esculentus var. Kiran and A. moschatus x A. esculentus var. Anakomban could not be cultured in vitro as the embryos aborted by the 11th day after pollination and those less than 10 days old could not be distinguished and excised.
Multiple shoot production of embryo derived plants was accomplished when cultured on MS supplemented with BA 1 mg 1-1 and CW 150 ml 1-1. Rooting of in vitro produced plants was achieved in Ms medium supplemented with IAA 1 mg 1-1.
Plants of the crosses A. esculentus var. Kiran x A. moschatus and A. esculentus var. Anacomban x A. moschatus showed an ex vitro survival percentage of 36.00 and 30.00 respectively. Preliminary graft transmission study using the hybrid plants as scion revealed their resistance YVM.
The study projects the possibility of using the in vitro rescued interspecific hybrids of A. esculentus var. Kiran x A. moschatus and A. esculentus var. Anakomban x A. moschatus for future breeding works, to produce A. esculentus lines with high yield, quality and resistance to yellow vein mosaic virus.
