Summary, etc. | The studies on in vitro shoot tip culture of banana for small scale production system were carried out in the Plant tissue culture laboratory, Department of Pomology and Floriculture, College of Horticulture, Vellanikkara.Explants collected during March-April recorded better survival percentage. Filter water (micro carbon filter) and sterilized filter water can be used as an alternative to distilled water tor the preparation of the media. Surface sterilizing the explants with mercuric chloride 0.1 per cent for 10 minutes recorded hundred per cent survival. For culture establishment, half MS +BA 3.0mg r' +NAA 1.0 mg rl was found to be an effective alternative. Half strength of MS was on par with full MS in terms of multiple shoot producton. Addition of half tablet of vitamin B complex resulted in better multiple shoot production. Table sugar at 2.0 per cent and at 3.0 per cent was found to be an effective alternative to sucrose as carbon sources. Agar 0.6 per cent or agar 0.5 per cent with gelatin 0.3 per cent was found to be good gelling agent similar to that of agar 0.7 per cent. Coconut water 20 per cent +BA 2.0 mg r' and palmyrah liquid endosperm 150 g r' + BA 2.0 mg r' were found to be the best alternative to BA, whereas triacontanol 2.0 ml rl was the best substitute for NAA. Coconut juice 20 per cent, cucumber juice 25 per cent, coconut milk 15 per cent and 20 per cent and casein hydrolysate 100 g I -I were found to be the effective medium supplements. The growth and development of shoot tip cultures in MS basal medium supplemented with 80 per cent v/v cyanobacterial extracts, enhanced more multiple shoot formation. For in vitro. rooting half MS in combination with NAA 2.5 mg rl produced maximum number and length of roots. For in vitro rooting, half tablet of vitamin B complex was found to be the best substitute for the vitamins. Table sugar 3.0 per cent, 2.0 per cent and palmyrah liquid endosperm 150g rl+ table sugar 1.5 per cent was found to be an effective carbon source in in vitro rooting medium. Agar 0.6 per cent and 0.5 per cent in combination with gelatin 0.3 per cent was found to be good substitute for standardized gelling agent (agar 0.7%). The highest number of roots (17.66) was produced by the plantlets grown in media supplemented with 1.0 mg r' triazoles, compared with 12.55 in control plants. Triazole concentrations above 2.0 mg rl resulted in the production of a large number of very short, thick roots. For ex vitro establishment of plantlets, vermiculite was found to be the best rooting medium. Better establishment of plantlets was achieved in goat manure+cow dung +sand mixture (1: 1: 1). Weekly spray of 17:17:17 (N: P: K complex) 1.0 per cent solution resulted in maximum number of leaves, length and width of leaves and height of plantlets. The expenditure involved in production of a single plantlet in cost effective method was Rs. 6.83 whereas for the same in conventional method it was Rs.l1.90 thereby reducing the cost per plantlet by Rs.5.07.
