Item Efficacy of Intrauterine Administration of Gentamicin in the Treatment of Clinical Endometritis in Crossbred Cattle(Department of Animal Reproduction, College of Veterinary and Animal Sciences,Mannuthy, 1985) Varadarajan, M; Prabhakaran Nair, P KThe objective of the study was to assess the efficacy of intrauterine infusion of Gentamicin, a bread spectrum antibiotic with deep tissue penetrance either alone or in combination with Stillboestrol in the treatment of clinical endometritis. Isolation of the infective organisms and sensitivity test were also carried out on a limited number of cases. One hundred and seventy crossbred cows and heifer with clinical endomtritis formed the materials for the study. These animals were randomly allotted into two treatment groups. Group 1 comprising of 86 animals were given 250 mg of Gentamicin intrauterine and group 11 comprising 250 mg of Gentamicin intrauterine and group 11 comprising of 84 animals were given a combination of Gentamicin and 10 mg of Stilboestrol. Those animals which were completely free of infection were inseminated in the next heat and rest were given a second infusion of the above drugs. Conception rate was worked out both on 90 days non return rate and actual pregnancy verification. Uterine discharges from 25 animals were collected for bacterial isolation and sensitivity test. Among 8352 cattle examined during the period of study 807 (9.66%) were found to be affected with clinical endometritis. Eighty six cases treated with Gentamicin gave a single insemination conception rate of 56.45 per cent with an overall conception rate of 72.58 per cent needing 1.91 inseminations per conception. On the ether hand, 84 cases receiving Gentamicin, Stilboestrol combination recorded a single insemination conception percentage of 40.90 with an overall conception of 63.63 per cent requiring 2.39 insemination per conception. There was no variation between groups with respect to percentage conception and average number of days from treatment to conception. The percentage clinical recovery with single infusion in group 1 and 11 were found to be 96.51 and 94.03. The number of intrauterine infusion was not found to influence the conception rate and the recovery period. The study also revealed that the parity of the animal in both the treatment groups did not influence the effectiveness of the treatment with respect to percentage conception and the average number of days from treatment to conception. It can be concluded that Gentamicin or Gentamicin Stillboestrol combination is equally effective in the treatment of clinical endomtritis. However, addition of Stillboestrol in the Gentamicin infusion did not have any added beneficial effect. The bacterial isolates were Haemophilus (8%), Staphylococcus (20%), Bacillus (4%),Pasteurella (8%) Actinobacillus (4%), Heromonas (12%), Lectobacillus (4%)and Streptobacillus (4%). The sensitivity of the isolates to Gentamicin, Septran, Chloramphenicol, Tetracycline and Ampicillin were respectively 60, 44, 28, 28 and 8 per cent. Isolation and sensitivity study has proved that most of the isolates were sensitive to Gentamicin and hence it is considered to be a very effective antibiotic for treating uterine infection.Item Chromosome profile of Indian elephants ( Elephas maximus indicus)(Department of Animal Breeding and Genetics, College of Veterinary and Animal Sciences, Mannuthy, 1988) Sakthi Kumar, A; Mukundan, GA karyological study in Indian elephants using peripheral venous blood collected from 7 tuskers, 7 cow elephants and one makhna belonging to some temples and private owners of Trichur District was undertaken with objectives of a) to evolve a technique for chromosome studies of elephants. b) to find out the chromosome number in Indian elephants, and c) to suggest sex chromosome mechanism in sex determination. Standardization of technique Comparison of efficacy of phytohaemagglutinin – M (Difco) and Poke – weed mitogen (GIBCO), in two different culture media namely TC 199 and RPMI – 1640, as mitogens showed that phyto – haemagglutinin – M yielded a mitotic index of 0.33 and 0.66 per cent in medium TC 199 and RPMI – 1640 respectively; poke – weed mitogen yielded a mitotic index of 5.33 and 5.66 per cent respectively in TC 199 and RPMI – 1640. Statistical analysis revealed that the efficacy of the two different media used was not significantly different, whereas the efficacy of two mitogens differed significantly at five per cent level. Poke – weed mitogen was found to be better than phytohaemagglutinin – M as far as indusing mitoses in elephant lymphocyte cultures over a 72 hour culture period was concerned. Colchicine treatment using 0.1 ml colchicine solution (0.0001%) for a period of one hour yielded satisfactory chromosome preparations. Karyotype The karyotype of the tusker, cow elephant and makhna revealed a diploid chromosome number of 2n = 56, comprising of 54 autosomes and 2 sex chromosomes. The autosomes were classified into 6 submetacentric and 21 acrocentric chromosomes. The X – chromosome was a submetacentric in all the three groups of elephants whereas the Y – chromosome was a small acrocentric in the tusker as well as the makhna. Analysis of relative length of chromosomes showed that the largest chromosome pair measured a relative length of 6.973 per cent and the smallest chromosome was the Y – chromosome measuring a relative length of 1.710 per cent. Position of centromere on the basis of centromeric index suggested that the centromere was farthest from the centre of the chromosome in the second autosome pair and nearest to the centre in the X – chromosome among the submetacentric chromosomes. Neither autosomes nor sex chromosomes possessed satellites on karyological examination. The elephants studied exhibited XX/XY sex chromosome mechanism. The cow elephant possessed XX sex chromosome complement while both tusker and makhna possessed XY sex chromosome complement. A comparison between the karyotypes of tusker and makhna did not show any variation in (a) basic number, (b) relative length (C) position of centromere, (d) absence of satellites on the chromosomes and (e) the sex chromosome complement. The chicks were fed broiler starter from 0-6 weeks and broiler finisher from 7-8weeks. The additives were incorporated at levels recommended by the manufactures and the duration of the study was for 8 weeks
